■ Title: The Role of Oxysterol-Binding Protein-Related Family proteins in the Anticancer Activity of Steroidal Saponin OSW-1

■ Abstract: My research is focused on a naturally occurring anticancer compound named OSW-1. Although OSW-1 is known to be very effective for killing cancer cells but their target(s) as well as their mechanism to kill cancer cells are still being observed. Identifying cancer drug target(s) and understanding their mechanism of actions become the major attention in drug development to further enhance the efficacy and the effectivity of the drug for cancer treatment. Without comprehensive understanding of the drug’s functions, target(s), and mechanisms, a particular drug cannot be subjected for further drug development. In this presentation, we analyzed the OSW-1 binding activity using Cellular Thermal Shift Assay (CETSA) and functional relationship of oxysterol-binding protein-related protein family (ORPs) member named OSBP and ORP4 that were presumed to be the target of OSW-1 anticancer activity but their endogenous binding and their mechanism of action with OSW-1 remain unclear.
